Developing New Molecular Diagnostic Tools for Leishmaniases, Trypanosomiases, Relapsing Tick Fever and Tuberculosis

The Infectious Diseases and Tropical Medicine Unit (IDTMU) aims to maintain its role as a national centre of excellence in tropical medicine and in microbiological parasitological diagnostic services in Palestine. It is committed to developing further its national role through collaborative networking in order to offer its expertise to the medical community and serve the beneficiaries of the Palestinian health system.
 

Research done by the IDTMU at the Institute stems from field questions and observations, generates knowledge on the biology of pathogens and vectors and leads to the development of appropriate tools for detection and typing of pathogens and vectors in relation with:

  • disease control activities;
  • epidemiology to the molecular level
  • Transmission, vectors and reservoirs;
  • important phenotypes (pathogenicity , virulence and drug susceptibility);
  • vector parasite interaction and bio-communication.

We only mention a summary of these activities, to demonstrate the magnitude of the expertise accumulated by the IDTMU.

The following tools have been developed within the network, and are accessible (training and equipment) for all members of the network:

  • Diagnosis and molecular genotyping: Leishmania, Trypanosoma spp., Mycobacterium; and Borrelia
  • Optimal procedures for DNA extraction from host tissues (insects, blood, archive clinical samples and biopsies) and environmental sources;
  • Single PCR: Leishmania (rDNA, kDNA, and 7 SL genes), Trypanosoma sp. (VSG and rDNA genes), Mycobacterium (narGHIJ promoter, oxyR, 3’end of RD1, rpoB genes, repetitive sequence IS6110); and Borrelia (flagelin gene).
  • High resolution melt (HRM) PCR: Leishmania (rDNA, kDNA, and 7 SL genes), Trypanosoma sp. (VSG and rDNA), Mycobacterium, and Borrelia.
  • ITS PCR and sequencing: Leishmania sp. and Trypanosoma sp.;
  •  PCR-RFLP: Leishmania (ITS1, 7SL genes)
  •  Serology for Leishmaniasis (ELISA);
  •  RT-PCR: Leishmania (7SL gene) and Mycobacterium for treatment follow up;
  •  Microsatellite analysis for population genetic on Leishmania major, L. tropica, L. infantum, and phlebotomus papatasi as vector for L. major.
